Mr. Big from Narc coin-op is impossible to beat without a partner, and the game itself is extremely difficult to beat. You need at least a $20 of quarters to get to him.
It is literally impossible to beat without a partner. Once you die and you put in another quarter, the level resets and you're up against Mr. Big with full life yet again. You really need to hit him from both sides or else he just shoots you or rams the bejesus out of you. He does ridiculous amounts of damage to you. So not even the Bagelmeister would be able to do it.
You said that about 1943? You should play NARC in Full Mode and try beating Mr. Big. I know if I play 1943 with unlimited quarters I'll eventually finish the game. Don't get me wrong, I totally respect WB's gaming prowess, but Narc coin-op is unbeatable without a partner.

That sounds like a tough fight. I have played NARC very little, but that sounds really challenging.
Actually I remember asking the person who submitted it if they could even beat it. The answer I got was basically "lol I dunno but we'll see". That's why it was so annoying.
Oh he is a beast for sure, and like Joz said if you lose and have to continue any damage you did to him is nullified. You have to do it in one credit to win.
As for hardest bosses? Only one that comes to mind atm is Jin Pachi from Tekken5. He had an unblockable move and the camera angle was intentionally different from the rest of the game. The camera angle thing was infuriating and it made him much harder than he should've been. All this could also be because I'm not uber pro at Tekken though.
Yeah I remember sidestepping it but the way it launched was very annoying and hard for me to time. I usually played up to him and just stopped if he killed me once or twice. I hate giving up but when a boss is intentionally different from the rest of the game it really turns me off.
I get frustrated easily by artificial difficulty. I like when bosses do everything the player can do, but better. Then it actually seems like a test of skill not luck.
